چکیده

Polysaccharides and related glycopolymers offer a unique challenge to analytical ultracentrifugafion for two clear reasons. Firstly, there is a growing demand for basic solution biophysical information on these substances: molecular weights, molecular weight distributions, conformation, conformation distributions; information which analytical ultracentrifuge can, in principle, supply. This information is important for the understanding of for example the performance of so-called "commercial,, polysaccharides in the food, drug and oil industries and for understanding the behaviour of glycopolymers such as mucus glycoproteins in health and disease.

Challenges for the modern analytical ultracentrifuge analysis of polysaccharides.

This article reviews some of the recent advances in analytical ultracentrifugation and how these advances have impacted--and can impact--on our understanding of the size, shape through conformation modelling, interactions and charge properties of polysaccharides in solution, particularly when used in combination with other solution techniques and also imaging techniques. Specifically we look at...

Solution conformation and flexibility of capsular polysaccharides from Neisseria meningitidis and glycoconjugates with the tetanus toxoid protein

The structural integrity of meningococcal native, micro-fluidized and activated capsular polysaccharides and their glycoconjugates - in the form most relevant to their potential use as vaccines (dilute solution) - have been investigated with respect to their homogeneity, conformation and flexibility. Sedimentation velocity analysis showed that the polysaccharide size distributions were generall...

Harding, Stephen E. and Gillis, Richard B. and Adams, Gary G. (2016) Assessing sedimentation equilibrium profiles in analytical ultracentrifugation experiments on macromolecules: from simple average molecular weight analysis to molecular weight distribution and interaction

Molecular weights (molar masses), molecular weight distributions, dissociation constants and other interaction parameters are fundamental characteristics of proteins, nucleic acids, polysaccharides and glycoconjugates in solution. Sedimentation equilibrium analytical ultracentrifugation provides a powerful method with no supplementary immobilization, columns or membranes required. It is a parti...

CHAPTER 12 Analysis of Polysaccharide Size , Shape and Interactions

In the earlier Royal Society of Chemistry book Analytical Ultracentrifugation in Biochemistry and Polymer Science,we reviewed the progress and potential of the ultracentrifuge for providing fundamental information about polysaccharides in what for many is their natural state in solution.r Our chapter was reinforced by contributions from Lavrenko and co-workers,2 who looked in detail at the conc...
